This is one of those questions that has more than one correct answer: It is easy if you have the necessary tools and experience; but quite a challenge for a beginner. To minimise fluid loss you can either clamp the flexible hose (frowned on by some) or buy some brake pipe and connecting parts and make yourself a blanking piece to screw in. You'll still need to bleed the brakes after refitting. If you are only looking to paint the caliper without changing the discs and pads at the same time then I'd recommend you just clean up and mask off very carefully and thoroughly and paint in-situ. In all honesty it doesn't need to be perfect because you can't see the caliper very clearly through the wheel anyway and your beautifully painted caliper soon gets a bit dirty.
